Bisulfite sequencing analysis of IG-DMR (CG4 and CG6) and MEG3-DMR (CG7) using DNA samples from human parthenogenetic and fertilized embryonic stem cells. (A) Schematics of IG-DMR and MEG3-DMR in Dlk1-Dio3 imprinting clusters. Normally, CG4 and CG6 structures from IG-DMR and CG7 from MEG3-DMR were methylated (black dots) in paternally derived chromosomes, and unmethylated (white dots) in maternally derived chromosomes; BGHPES-1 (B) and -2 (C) and control FHES-Cont (D) data. Each horizontal line indicates a single subcloned allele. Hypomethylation was observed in BGHPES-1 and -2, and balanced methylation in control cells. IG-DMR, intergenic differentially methylated region.
