Effects of recombinant endothelin-1 (ET1) on regulation of pre-differentiated human mesenchymal stem cells (hMSCs). (A) Proliferation of hMSCs treated with or without different doses of ET1 was analyzed by quantifying total DNA content. (B) Relative mRNA expression levels of potency- and lineage-related markers in hMSCs treated with or without different doses of ET1 for two passages were analyzed by quantitative RT-PCR. *P < 0.05; n = 3. (C) Growth curves of hMSCs treated with or without 0.1 μM ET1 were generated by calculating cumulative population doublings. (D) Expression of surface antigens of hMSCs treated with or without 0.1 μM ET1 for two passages was assessed by flow cytometry.
