Rapamycin attenuates bafilomycin A1-induced iPSC death. iPSCs were treated with 0 (a), 5 nM (b), 50 nM (c), and 100 nM (d, e) of bafilomycin for 24 hours (ad) or 48 hours (e). Bafilomycin A1 induced cell death in a concentration-dependent manner (ad), leading to substantial cell loss after 48 hours at 100 nM (d). Addition of 200 nM rapamycin attenuated bafilomycin A1-induced cell death and maintained iPSC normal cell density (f, g). (h) Anti-ACTIN and Anti-LC3B immunoblotting were carried out with iPSC lysates from control iPSCs (Con), or 24 hours of treatment with 200 nm of rapamycin (Rap), or with 100 nM of bafilomycin A1 (Baf), or with both (Rap + Baf).
