The TSCM are transferred into LCL-burden mice for stem and memory T cell behavior. a b Adoptive transfer protocol of the TSCM. On day − 3, irradiated mice were inoculated with E007 or E001 cells to set up the LCL-burden mouse model. On day 0, E007-specific TSCM or TEM and TEF (TEM + EF) were transferred into the mice intravenously (iv). Blood sampling was taken once a week for 5 weeks after the T cell treatment. On day 35, the mice were sacrificed for detection of human T cells and LCL cells in spleen and bone marrow. c–e The prepared TSCM cells had superior persistence in vivo. The frequency of human CD3 + CD8+ T cells (c), the frequency of TSCM cells (CD45RA+ CD62L+) in human CD3+ CD8+ T cells (d), and the absolute number of TSCM cells in 50 μl blood (e) were detected by FCM. Data are represented as mean ± SD of five mice. (*p < 0.05; NS, p > 0.05)
