miR-218 targets hypertrophy-related genes in MSC. The miR pulldown assay was done in MSC (N = 2 donors). After cell lysis, biotinylated miR-218 mimic (gray) or a control non-targeting miR mimic (NC, black) was incubated with cytosolic fractions of the MSC cell extract, then immobilized and purified on magnetic streptavidin beads. After elution and total RNA extraction, abundancies of pulled-down mRNAs for indicated genes were measured using qRT-PCR and normalized to a housekeeping gene (HPRT).
