The inhibition of mtROS production mediates the suppression of NLRP3 inflammasome via ADSCs. Cells were pretreated with Propionibacterium acnes at MOI = 10 for 24 h and co-cultured with ADSCs in a Transwell system. The ROS inhibitor NAC (10 mM) was added 1 h before the addition of P. acnes. A Intracellular ROS stained in green fluorescent DCF was observed under a fluorescence microscope. B Fluorescence microscopy images of ASC (red). The nuclei were stained with DAPI (blue). The arrows indicate ASC specks. C NLRP3, pro-IL-1β, and pro-caspase-1 in cell extracts (CE) and IL-1β and caspase-1 (p10) in supernatants (SN) were determined by western blot.
