Cryopreserved MSC overexpressing CD::UPRT::GFP exhibited high recovery and prolonged therapeutic gene expression. (A) Cryopreserved modified or freshly modified cells were replated onto 6-well plates (n = 3). One day later, adherent cells were harvested and enumerated. Percentage recovery was calculated using the number of adherent cells to number of cells seeded into each well. (B) Cryopreserved MSC overexpressing CD::UPRT::GFP were replated at 1,000,000 cells/well in the 6-well plate and grown in culture over a period of 5 days. On day 1, 3 and 5 post replating, RFU was measured. On day 5, the cells were harvested from each well and the transfection efficiency was quantified by FACS. Data of biological triplicates (n = 3) were presented as mean ± SD. Representative image from each condition is presented
