From: Roles of microRNA-34a targeting SIRT1 in mesenchymal stem cells

Fig. 4

miR-34a exerts pro-apoptotic effects via activation of the mitochondrial apoptosis pathway. MSCs were transfected with siRNA-NT, siRNA-SIRT1, miR-34a inhibitor, or siRNA-SIRT1 cotransfected with miR-34a inhibitor under normal and H/SD conditions (MSCs were transfected for 72 hours and exposure to H/SD and maintained as such for 6 hours). Then ∆Ψm (a) was analyzed by measuring JC-1 fluorescence, and cytosolic and mitochondrial cytochrome c expression (b, c) were measured with western blot. Each column represents mean ± SD from three independent experiments. *P <0.05 vs. H/SD scramble, â–³P <0.05 vs. miR-34a inhibitor, â–²P <0.05 vs. siRNA-SIRT1. H/SD hypoxia and serum deprivation, miRNA microRNA, SIRT1 silent information regulator 1, siRNA small interfering RNA
