The effect of selection on survival of RW4 and Sim1-Puro embryonic stem cells (ESCs). a Diagram depicting induction and selection process. Induction and differentiation nomenclature is indicated by parenthesis while the time course (in days) is denoted by the time line. Briefly, ESCs are allowed to aggregate for 2 days (2−) to form embryoid bodies (EBs). EBs are induced with retinoic acid (RA) and sonic hedgehog (Shh) for 5 days (5+) and selected with puromycin in the presence of RA and Shh for 1 day (6+/ 1p).
