Effect of H2O2 on apoptosis in BMSCs. BMSCs were injured with H2O2 at different concentrations from 200 to 800 μM for 24 h, and the a cell viability (scale bar = 100 μm), b LDH concentration, and c degree of apoptosis indicated by TUNEL+ (scale bar = 50 μm) and d Annexin V+/PI+-labeled cells were then determined. Nuclei were labeled with DAPI. Moreover, BMSCs were exposed to H2O2 for 24 h, and cell lysates were collected and subjected to western blot detection of the Bcl2, Bax, and C-Casp-3 proteins, with normalization to β-actin. Cells treated with PBS served as a control group. Values are expressed as the mean ± SD (n = 3 independent experiments). *p < 0.05 compared with the control group. One-way ANOVA followed by the LSD test was used to analyze the significance of differences. Morphological images are shown at the original magnification.
