Notch signaling is activated and required for the generation of human MuRC in vitro. A Total mRNA were isolated from human myoblasts in growth medium (MB), 48Ā h MuRC and 96Ā h MuRC. qRT-PCR of Pax7, MyoD and the Notch target gene, Hes1. Results are presented as meanā€‰Ā±ā€‰SEM.; nā€‰=ā€‰6, one-way ANOVA. B Total protein extracts were isolated from human myoblasts in growth medium (MB) or after 24Ā h in DM. MuRC and myotubes fractions were separated after 48Ā h, 72Ā h or 96Ā h in DM. Western blot analysis for Pax7, MyoD and the notch intra-cellular domain (NICD). Results are presented as meanā€‰Ā±ā€‰SEM for myoblasts (MB), 48Ā h MuRC and 96Ā h MuRC; nā€‰=ā€‰6, one-way ANOVA. Full-length blots are presented in Additional file 1: Fig.Ā S1 C Immunofluorescence staining for Pax7 (green), MyoD (red) and DAPI (blue) of human myogenic culture after 48Ā h in differentiation medium (DM) in the presence of DMSO or the Notch inhibitor, DAPT. The images shown are representative of the three independent experiments. Results are presented as meanā€‰Ā±ā€‰SEM; nā€‰=ā€‰3 with 3 images analyzed per experiment, unpaired t test. Scale bar: 10Ā Ī¼m
