Fig. 4

Transcriptional profiles of human Pax7High and Pax7Low MuRC subpopulations. All human cells (myoblasts, MuRC-Pax7High and MuRC-Pax7Low) were fixed/permeabilized and stained for Pax7 before flow cytometry and mRNA isolation. A Biological samples were represented by principal component analysis (PCA). PC1/PC2 analysis demonstrated that human MuRC and human myoblasts displayed very different expression profiles (PC1), whereas for each biological sample analyzed by RNA-seq, we observed that Pax7High and Pax7Low MuRC are very similar in gene expression profile. B Volcano plot of the supervised analysis comparing myoblasts and Pax7High MuRC and comparing Pax7High MuRC and Pax7Low MuRC transcriptomes. Strong transcript modifications between myoblast and Pax7High MuRC were observed with 3393 modified genes whereas the changes were moderate between Pax7High MuRC and Pax7Low MuRC with 103 modified genes (Log2FCā€‰>ā€‰0.5, Pā€‰<ā€‰0.01). C Heatmap showing the number of normalized reads of MuRC-RNA-seq data for the 103 modified genes between Pax7High MuRC and Pax7Low MuRC populations. (D, E) Heatmap showing the average number of normalized reads of myoblasts (nā€‰=ā€‰5), of Pax7High MuRC (nā€‰=ā€‰6) and of Pax7Low MuRC (nā€‰=ā€‰6) RNA-seq data for a selection of genes
