Probing HFP-MSC cell lysates before and after 3D bioreactor culture senescence or apoptosis markers to rule out activation of these cellular processes during EV collection. (A) Analysis of p21 expression before (CTRL) and after EV collection following treatment of HFP-MSCs with the indicated blood product (FCS, CPRP, hypACT). Lysates of HFP-MSCs treated in 2D with 5 µM etoposide or 30 µM H2O2 for 48 h and 2 h, respectively, were used as positive controls. (B) Analysis of caspase 3 cleavage before (CTRL) and after EV collection following treatment of HFP-MSCs with the indicated blood product (FCS, CPRP, hypACT).
