Lipid morphology as a function of oxygen levels and cytoskeletal inhibition. Oil Red O staining (red) qualitatively demonstrated changes in lipid accumulation in adipose-derived stem cells (ASCs) that had been differentiated for 21 days in 20% O2 and 5% O2 conditions. ASCs under 20% O2 conditions without cytoskeleton inhibitor treatment (A) appeared to have a higher percentage of Oil Red O positive cells compared to ASCs under 5% O2 conditions without inhibitor treatment (D). The addition of either cytoskeletal inhibitor, cytochalasin D (B,E) or blebbistatin (C,F), appeared to increase the percentage of lipid stained cells regardless of oxygen conditions compared to non-inhibited ASCs (A,D). ASC nuclei were counterstained with Hoechst (blue). Scale bar = 30 μm.
